  • FiltrationFiltration is certainly one of the oldest separation techniques although its real origins are lost in the depths of time. The Arabicalchemists knew about distillation, extraction and filtration. In the 13th centrury the ”Summa perfectionis magisterii“ appeared underthe pseudonym ”Geber“; the author was probably the Franciscan Paulus of Taranto. In a factual form which is very unusual for thisperiod he describes alchemy mainly from a scientific point of view and explains the construction and use of filtering instruments andmany other typical laboratory devices still in use today.

    MembranesThe word membrane is coming from the latin word ”membrana“, which means skin. Today we understand the term Membrane as a”skin“ with semi permeable functions. From the first membrane in use, we know that they were produced from natural materials likeanimal skin and natural fibers. That permeability is also found in natural organs like pig, fish urinary bladder and intestine. All of themshow a permeability to certain compounds. The microscopist R. Cook describes for the first time results on Membrane technique byreporting about toward the porosity of cork (1667). The phenomenon of osmosis is describe by Abbe Nollet in an essay (1748) wherehe speaks about the characteristics of animal bladders. On the nineteenth century was the beginning of the systematics analysis formembrane separation techniques.

    Membrane-Butler

    Membrane filters for microbiologicalchecks must be carefully handled toensure that they remain sterile andthat quantitative results are beeingobtained.

    The Membrane-Butler allows packagesto be opened quickly and in this wayensures the optimal handling of allMicroPlus and ME membrane filters.Membranes suitable for Butler systemsare designated as STL (see page 21 for

    ordering information). The dispenserbox is inserted in the Membrane-Butler,the sterile packaging is placed in theroller system and that’s it. Electroniccontrols are used to remove amembrane filter from its sterile packa-ging. The filter is then simply graspedwith a pair of forceps and can be usedfor filtration.

    The MBS I is a filtration system forhigh throughput laboratories workingin the area of microbiological qualitycontrol. It is of particular benefit forserial samples and users who havemore than 25 samples a day.

    The MBS I system consists of an elec-trical membrane-dispenser, a funneldispenser and a vacuum manifold. Thevacuum manifold is a modular systemwith 2-place modules which can beeasily connected to 4- or 6-place mani-folds. The system is set up to yourworking procedure and supports theergonomic handling of the membranefiltration step.

    MBS IMicrobiological Filtration System

    MBS I – Bringing a system into your qualitycontrol.

    When taking a fresh funnel from theMBS I, the Membrane-Butler E automa-tically dispenses a membrane from thesterile packaging. Thus special mem-branes in a leporello folded format(STL) ideally complement the system.Users working with two different mem-brane types (i.e 0.45 and 0.8 µm) canequip the system with a Butler Tower oftwo Membrane-Butler E units stackedtogether.The special sealing technique guaran-tees easy handling and a good integrityof the funnel and membrane duringfiltration. This reduces any cross-conta-mination to a minimum. Compared tostandard 250 ml funnels, the MBS Ifunnel can hold a 40 % larger volumewhich is especially convenient whenhandling foaming liquids. When usingthe MBS I system, sterilizing funnels orthe filter base in between filtrationsteps can be avoided. Investigationsrevealed time savings of up to 50 %.Also, the membrane filtration method isnow open for validation. Flaming is

    Schleicher & Schuell MicroSciencedeveloped the MBS II specifically formicrobiological quality control in thepharmaceutical industry.

    Handling of the system is particularlypractical with the filtration funnel andmembrane combined to form a ready-to-use sterile filtration unit. Duringfiltration the sample is protected by avented lid. When filtration is finished,the membrane is transferred to an agarplate or a Petri dish. Easy handling of the MBS II systemensures reproducible results and inaddition to that the system offers lessdanger of cross-contamination.

    MibScreen Bac and MibScreen Yeast

    MibScreen Kits are fluorescence kitsfor the rapid detection of micro-organisms.

    The MibScreen Kits are especiallydesigned to meet the requirements offastidious microbiological tests for pro-duct release in the beverage industry.Two kits are available one for bacterial-and a second one for yeast detection.

    After filtration through special micro-sieves 0.45 or 1.2 µm pore size the co-loring reagents are added to theresidues on the filter. If bacteria oryeast are present the reagents speci-fically indicate living or dead cells bylight source. Thus viable cells likePediococci, Lactobacilli or yeast will beeasily detected. The test method issecure and gives reproducible results.

    Microsieve Technology A novel approach to MicrobiologyQuality Control

    The Schleicher & Schuell MicroSciencemicrosieve is a new tool for rapid iden-tification and quantification of micro-organisms. The microsieve is made with high-techmanufacturing methods originated fromthe semiconductor industry. Micro-sieves are sieving membranes withperfectly controlled uniform pores insize of 0.45 and 1.2 micron. The microsieve is perfectly (optically)flat giving a sharp microscopic imageat all magnifications. The microsieve ismade of a non-polymeric material andis totally free from UV back lightscattering noise.

    MibScreen Bac

    Bac I/Yeast I-Solution are non-polar, non-fluorescent reagent which can pass through the cellmembrane whereupon intracellular esterases cleave off the diacetate group producing the highlyfluorescent compound. The compound will accumulate in cells which possess intact mem-branes so the green fluorescence can be used as a marker of cell viability. Cells which do notpossess an intact cell membrane or an active metabolism may not accumulate the fluorescentproduct and therefore do not exhibit green fluorescence. This may be used in combination withBac II/Yeast II-Solution staining as the non-viable cells will take up the Bac II/Yeast II-Solutionand stain red whereas viable cells will not take up the Bac II-Solution and should only staingreen. This 2-colour separation of non-viable and viable cells provides for a more accuratequantitation of cell viability than single colour analysis.Bac II/Yeast II-Solution is a non-permanent dye that can penetrate the membranes ofdying/dead cells. It intercalates into the major groove of the DNA and produces a highlyfluorescent adduct so non-viable cells can be identified by a red fluorescence.

    The stainless steel manifold for 3 or 6filtration units is fitted with stainlesssteel funnels. The apparatus can beautoclaved and sterilized by dry heatup to 180º C. These manifolds are onlysuitable for vacuum filtration. The areaof use is multipurpose: Microbiologicalquality control, residue analysis andserial filtration can be carried outrapidly and easily with a commondrainage outlet. Each unit has its ownseparate stopcock.
