Inhalt:
An

Alexandra Schieweck, Tunga Salthammer
In seiner Konzeption

Inhalt englisch:
Introduction The German initiative to develop new human biomonitoring (HBM) methods
forsubstances of interest is a cooperation of the Federal Ministry for the Environment,
Nature Conservation,Building and Nuclear Safety (BMUB) and the German Chemical
Industry Association (VCI) which started in2010. Emphasis is placed on substances either
with potential health relevance or for which an exposure of the
general population can be assumed. Methods After identification of relevant chemicals by
an expert panel HBMmethods are developed and first applied to about 40 non
occupationally exposed persons. The analyticalmethods are cross-validated by the working
group ´Analyses in biological Materials of the German ResearchFoundation (DFG) and
published together with pilot-test results in international, peer-reviewed journals.
ResultsNew HBM methods for methylenediphenyl diisocyanate (MDI ), used in
polyurethane and adhesives, thevulcanization accelerator 2-mercaptobenzothiazole (2-
MBT), the plasticizer alternatives Hexamoll® DINCH anddi-2-propylheptyl phthalate
(DPHP), the alkylphenols 4-tert-octylphenol and 4-nonylphenol and the technicalsolvents
N-methyl- and N-ethyl-2-pyrrolidone (NMP/NEP) have already been developed and applied.
For someof these chemicals a considerable background exposure seems to be unlikely
(e.g. MDI), for other substances(e.g. NMP/NEP) a ubiquitous background was detected. For
DINCH and DPHP, trends of increasing exposurewere observed in urine samples from the
German Environmental Specimen Bank, mirroring the marketintroduction of the
substances. The relevance of the findings needs to be assessed by toxicologically
derivedHuman Biomonitoring Values (Biomonitoring Equivalents) and by population
representative studies.Consequently, we plan to include these substances in the 5th
German Environmental Survey. The results willcontribute to a realistic exposure and risk
assessment for these "new” chemicals in German children.

Inhalt:
t/a CO2-Äquivalente.
t/a anstatt 1.400 t/a).

Inhalt englisch:
N-Methyl-2-pyrrolidone (NMP) and N-ethyl-2-pyrrolidone (NEP) are multi-purpose organic
solvents in industry. Both are developmental and teratogenic toxicants in rodents. NMP is
classified as a REACh substanceof very high concern. Because of their toxicological profile
and their broad application resulting in a possible exposure of the general population,
NMP and NEP were chosen as target substances for the cooperation project between the
German Federal Ministry for the Environment, Nature Conservation, Building and Nuclear
Safety (BMUB) and the German Chemical Industry Association (VCI) aiming to establish
human biomonitoring (HBM) methods for "new” substances of interest. NMP and NEP are
metabolized to 5-hydroxy-N-alkyl-2-pyrrolidones (5-HNMP, 5-HNEP) and 2-hydroxy-N-
alkylsuccinimides (2-HMSI, 2-HESI). We analyzed these specific metabolites in 24-hour
urine samples from the German Environmental Specimen Bank. For this purpose, 20
randomly selected samples collected in 1996 and in 2012, respectively, were analyzed by
a sensitive and specific GC-MS/MS method with isotope dilution quantification. We
detected NMP metabolites in100% and NEP metabolites in 95% of all samples. Despite the
considerable differences in the elimination halftimes of the alkyl pyrrolidone metabolites,
the correlations between the metabolites were rather strong (NMP: r=0.51; NEP: r=0.67).
An exposure determined through one metabolite is thus confirmed by the
other129metabolite. Median NMP metabolite levels were comparable between 1996 (5-
HNMP 50 ìg/L, 2-HMSI 46 ìg/L) and 2012 (5-HNMP 39 ìg/L, 2-HMSI 41 ìg/L). Surprisingly,
urinary levels of NEP metabolites were approx. 10times higher in 1996 (5-HNEP 14 ìg/L, 2-
HESI 42 ìg/L) as compared to 2012 (5-HNEP ~1 ìg/L, 2-HESI 5ìg/L). We would have
expected a reverse trend for NEP since NEP has only recently been introduced into the
market as a substitute for NMP. The sources of past and present exposures to NMP and NEP
warrant further investigations.

Europeans are still substantially exposed to chemicals. Toxicological and epidemiological
studies
have revealed several associations between exposure and health effects for numerous
substances. In Germany exposure to various chemicals is well documented by the German
Environmental Survey (GerES) and the Environmental Specimen Bank CESB). Many relevant
sources of exposure and chemicals of concern have changed over time: While in the past
ambient air and drinking water were particularly important, today's main sources are
nutrition, indoor air, cosmetics, and consumer products. After identification of substances
of toxicological relevance and to which the general population might considerably be
exposed new human biomonitoring (HBM) methods for i. a. the sensitizing agent
methylenediphenyldiisocyanate (MDI), the two plasticisers Hexamoll® DI NCH®,Di-2-
propylheptylphthalate CDPHP)- both substitutes for Di-2-ethylhexylphthalate (DEHP)- and
parabenes have been developed and-if appropriate- applied to ESB samples. While MDI
and DPHP seem not to be related with a measurable exposure of occupationally non-
exposed persons, DINCH levels increase constantly since marketing of this new substance.
The relevance of these findings is to be assessed by derivation of toxicological
Human-Biomonitoring-Values as well as by HBM studies in the population-representative
GerES V. lnvestigation of chemicals relevant today by HBM helps to prioritize further
toxicological investigations and provides guidance for consumer decisions and policy.
However, a continuous monitoring also of well known substances is essential for assessing
impacts of chemicals
on children. GerES and ESB are funded by the Federal Ministries for the Environment as
well as Education and Research.

The aim of this study was to investigate a possible time trend and status quo of
dipropylheptylphthalate (DPHP) exposure. DPHP is used as a substitute of other high
molecular weight phthalates in high temperature applications (e.g cable wires, roofing
membranes, etc.). DPHP was selected in the cooperation project between the German
Federal Ministry for Environment (BMU) and the German Chemical Industry Association
(VCI) due to its listing as High Production Volume (HPV) chemical in the European Union.
The BMU-VCI project establishes new human biomonitoring methods and biomarkers for
fifty emerging substances. 300 urine samples (24-hour voids) from the German
Environmental Specimen Bank were analyzed for three specific, secondary oxidized DPHP
metabolites (with hydroxyl, oxo and carboxy modifications of the alkyl side chain). Urine
samples were collected in the years 1999, 2003, 2006, 2009 and 2012, 60 samples per
year, from 30 male and 30 female volunteers (age: 21-29 years). The samples were
analyzed by liquid/liquid extraction followed by GC-HRMS, which enabled us to distinguish
between DPHP and Di-iso-decyl phthalate(DiDP) metabolites. The limit of quantification
was between 0.15 ìg/l and 0.3 ìg/l, depending on the metabolite. All samples were blinded
before analysis. DPHP metabolites were not detected in the years 1999-2006.Thereafter,
detection rates increased from 3.3% in 2009 to 21.7% in 2012. As expected, mono-oxo-
propylheptylphthalate (oxo-MPHP) was the most abundant metabolite, with concentrations
between <LOQ and 0.96 ìg/l.The daily DPHP intake was calculated via oxo-MPHP with a
maximum of 0.32 ìg/kg bodyweight/day. Our results show that DPHP exposure has
reached the general German population. However, exposure to DPHP inconsiderably lower
than for other high molecular weight phthalates. Future measurements will enable us to
monitor the development of DPHP exposure and advise risk management steps, if
warranted.

Introduction and methods: The ESB regularly collects human samples which are analyzed
for various
substances before being cryo-archived. Every year i. a. 24 h-urine and blood samples from
480 young adults (20-29 years of age) from four German cities are acquired. To document
time trends in human exposure to pesticides samples have been regularly analyzed for
Hexachlorobenzene (HCB) and Pentachlorophenol (PCP)since the 1980s. As initial
retrospective analysis 40 urine samples acquired in 1996 and 2012 have been analyzed for
N-(phosphonomethyl)glycine (Glyphosate). Results: ESB data reveal a major decrease in
average internal exposures to HCB (1985: 2.7 ìg/L vs. 2010: 0.09 ìg/L blood plasma) and
PCP (1985: 26.7 ìg/L vs.2010: 0.54 ìg/L blood plasma). Until the 1990s, HCB tends to be
higher in females. No substantial gender difference for this period was observed for PCP.
The internal Glyphosate exposure increased significantly between 1996 and 2012. The
fraction of quantifiable levels increased from 40% to 70%. On average the concentration
increased from below LOD (0.15 ìg/L) to 0.21 ìg/L urine. The highest Glyphosate value
measured in 2012 resulted in 0.65 ìg/L urine. Against expectation the concentration of
Glyphosates main metabolite Aminomethylphosphonic acid (AMPA) did not increase in
time. Also no significant correlation was observed between Glyphosate and AMPA levels.
Conclusions and Outlook: ESB data document the effect of banning PCP and HCB in
Germany in the 1980s. Concerning Glyphosate ESB measurements are in line with the
increasing application of this pesticide resulting in an increasing exposure of the
population. Levels measured in 2012 are well below toxicologically relevant
concentrations. However, due to the ongoing discussion on health effects of Glyphosate
further monitoring is warranted. Acknowledgements: The ESB is funded by the Federal
Ministry for the Environment, Nature Conservation, Building and Nuclear Safety (BMUB).

Introduction & methods: The German health-related environmental monitoring program
investigates
human exposures to environmental stressors and consists of the German Environmental
Survey (GerES) and the Environmental Specimen Bank (ESB). In addition to human
biomonitoring (HBM) and indoor monitoring for different segments of the general
population, standardized interviews performed in GerES yield reference data on exposure
influencing factors. The ESB obtains and cryo-preserves i. a. HBM samples, physiological
parameters, and self-administered questionnaire data on exposure-relevant behaviors
from adults (20-29 yrs.) on a yearly basis focusing on temporal changes. Results: Children
in GerES IV spent 15 h 31 min at home (4:46in other indoor environments, 3:43 outdoors)
differing by socio-economic status. Exposure models indicate the time spent at home being
less important for inhalative exposures than indoor pollutant levels. The ESB documents
how the decreasing fraction of participants with dental amalgam (1997: 83 % vs. 2013:10
%) is associated with decreasing urinary mercury (1997: 0.6 ìg/L vs. 2013: 0.07 ìg/L). ESB
data on decreasing 24 h urine volumes (1986: 1.3 L vs. 2013: 2.0 L) allows for adjusting
time trends in internal exposure. Conclusions &Outlook: GerES and ESB demonstrate how
exposure factors can be generated within regularly conducted177population studies. As
for all participants also pollutant monitoring data are available, individual associations
between exposure factors and internal exposures to various contaminants can be
investigated. These analyse scan also accounts for socio-demographic differences and
time trends in exposure relevant behaviors. The further development of both studies
includes online-questionnaires and interviews on potential sources of emerging pollutants.

The German Environmental Surveys (GerES) are large scale population studies repeatedly
been carried out since the mid-1980s. GerES I, II, and III focus mainly on adults, GerES IV
and GerES V (2014/2016) on children and adolescents. GerES evaluates pollutant body
burdens in population-representative samples, analyses the contribution of different
media (air, water, food) to the overall exposure, and links human biomonitoring (HBM) to
health data. GerES is conducted in cooperation with the National Health Interview and
Examination Surveys (NHIES) performed by the Robert Koch-Institute. The GerES V pilot
study was conducted in 2013 to validate instruments and field work procedures. GerES V
comprises HBM, indoor and drinking water monitoring, and standardized interviews,
tailored to exposure situation in Germany. GerES V focuses on emerging substances with
potential health relevance and/or assumed exposure of the general population (e.g.
plasticizer alternatives like Hexamoll® DINCH and di-2-propylheptyl phthalate (DPHP) or 4-
tert-octylphenol, 4-nonylphenol, the solvents N-methyl- and N-ethyl-2-pyrrolidone
(NMP/NEP), parabenes, and the vulcanization accelerator 2-mercaptobenzothiazole).
Exposure data on chemicals of concern already measured in GerES IV like phthalates and
other pollutants (i.a. metals, pesticides, PCB, cotinine) will be updated. The GerES V
concept demonstrates current strategies for elucidating environmental exposures. The
pilot study indicates comparatively low exposure to Bis (2-ethylhexyl) phthalate (DEHP).The
highest sum of the DEHP metabolites5OH- and 5oxo-MEHP measured in morning urine was
160 ìg/L which is well below the health-based guidance value. The main study will answer
the question whether a) plasticizer alternatives are nearly as omnipresent in children as in
a first small sample of non-occupationally exposed adults and b) screenings on emerging
chemicals in adults can advocate same analyses in studies on children and adolescents.

Introduction: A major objective of the European Environment and Health Action Plan 2004-
2010 was the EU wide comparison of the population's exposure to chemicals by human
biomonitoring (HBM). This required study harmonization. COPHES (Consortium to Perform
Human biomonitoring on a European Scale) systematically developed a harmonized
approach to conduct HBM on a European scale, an essential prerequisite to get comparable
data. Results: For all main aspects of study design and conduct alternatives were compared
and discussed in a transparent decision making process that can also be applied to other
studies. To reach acceptance of all involved partners, country specific aspects were always
considered. During workshops and by discussing written materials all partners brought up
their comments and became familiar with all instruments for fieldwork and methods for
quality assurance (e.g., fieldwork manual, questionnaires, and check lists). However, study
preconditions such as ethics requirements or acceptance of questionnaire components
differed between countries, giving a taste of the limits of harmonization. Finally, 17 of the
27COPHES countries conducted the DEMOCOPHES survey (DEMOnstration of a study to
Coordinate and Perform Human biomonitoring on a European Scale) using the harmonized
approach. Conclusions: The harmonized fieldwork instruments (basic questionnaire, urine
and hair sampling) turned out to be of high value for future HBM studies. Harmonization
requires detailed preparation but not necessarily in the same depth in each part of the
project. The challenge is to determine which procedures indispensably need perfect
harmonization and for which general guidelines are acceptable. Acknowledgements: We

There is mounting interest in the impact of the environment on childrens health and
development.
Recently, large-scale studies of childrens environmental health, including both
longitudinal and cross-sectional designs, have been planned and implemented in several
countries. These studies, aiming to better understand how social, environmental and
genetic factors affect childrens health; put special emphasis on the effect of environmental
stressors. Among a wide range of environmental stressors, the effect of chemical
contaminants is one of the focus areas to be investigated. In order to examine the
relationship between chemical exposure and childrens health in longitudinal studies,
individual exposure estimation is required rather than general information on the exposure
of the population. Exposure assessment utilizes monitoring, modelling and
questionnaires. Among these, biological monitoring (human biomonitoring) is one of the
most powerful methods to investigate individual exposure to chemical contaminants.
However, different studies employ different measurement methods, which hampers future
data pooling and analysis among the studies. France, Germany, Japan, Shanghai (China)
and the United States that are planning or conducting large-scale childrens environmental
health studies have gathered to form an international working group to harmonize outcome
and exposure measurement methodologies. The group identified three major contaminants
including mercury, 236phthalates and organophosphates for the first step of the
harmonization. The group started sharing information about sample collection, analytical
methods, quality assurance procedures and data reporting formats for the three
contaminants. Round-robin trials have also been conducted to identify key issues for
further harmonization. The progress of the working group discussion and trials will be
presented in the symposium.Acknowledgement: Environment and Childrens Health

Inhalt englisch:
Large scale multidisciplinary birth cohort studies are being designed and implemented in
several
countries around the world to examine the effects of the environment on childrens growth,
development and health. These studies feature investigation of behavioral, economic,
environmental, genetic and psychosocial factors on child growth and development with the
goal of better understanding how to improve the health of children. Study investigators will
collect environmental samples and biospecimens, and administer questionnaires, conduct
interviews, or make observations about household characteristics, lifestyle, medical care,
social interactions, and parents occupations. Investigators from several of the newer large-
scale birth cohort studies (planned or ongoing in China, France, Germany, Japan, France,
and the US) came together in2011 to form the Environment and Child Health International
Birth Cohort Group to coordinate efforts to harmonize core elements of their studies.
Harmonization provides an opportunity to share expertise and to collaborate in the
development of terminology, the selection of study visit measurements, and the collection
and analysis of data for exposures and outcomes of mutual interest. Results may lead to
the development of best practices and opportunities for additional international
cooperation. This presentation will describe the background of the Environment and Child
Health International Birth Cohort Group and outline its progress to date.

"neue" Weichmacher Hexamoll® DINCH® entsprechend seiner zunehmenden
verschiedenen Phthalaten hinzuweisen.

Inhalt englisch:
Within the European Environment and Health Action Plan an initiative to establish a
coherent human biomonitoring approach in Europe was started. The project COPHES
(COnsortium to Perform Human biomonitoring on a European Scale ) developed
recommendations for a harmonized conduct of a human biomonitoring (HBM) survey which
came into action as the pilot study DEMOCOPHES (DEMOnstration of a study to Coordinate
and Perform Human biomonitoring on a European Scale). Seventeen European countries
conducted a survey with harmonized instruments for, inter alia, recruitment, fieldwork and
sampling, in autumn/winter 2011/2012. Based on the countries' experiences of
conducting the pilot study, following lessons learnt were compiled: the harmonized
fieldwork instruments (basic questionnaire, urine and hair sampling) turned out to be very
valuable for future HBM surveys on the European scale. A school approach was favored by
most of the countries to recruit school-aged children according to the established
guidelines and country specific experiences. To avoid a low participation rate, intensive
communication with the involved institutions and possible participants proved to be
necessary. The communication material should also include information on exclusion
criteria and offered incentives. Telephone contact to the participants the day before
fieldwork during the survey can prevent the forgetting of appointments and first morning
urine samples. To achieve comparable results on the European scale, training of
interviewers in all issues of recruitment, fieldwork and sampling through information
material and training sessions is crucial. A survey involving many European countries
needs time for preparation and conduct. Materials for quality control prepared for all steps
of recruitment, fieldwork and sampling proved to be important to warrant reliable results.

Inhalt englisch:
Uranium occurs naturally in soil and rocks, and therefore where it is present in water-
soluble form it also occurs naturally in groundwater as well as in drinking water obtained
from groundwater. Animal studies suggest that the toxicity of uranium is mainly due to its
damage to kidney tubular cells following exposure to soluble uranium compounds. The
assessments of the absorption of uranium via the gastrointestinal tract vary, and this has
consequences for regulation, in particular the derivation of e.g. drinking water limit values.
Absorption rates vary according to the nature and solubility of the compound in which
uranium is presented to the test animals and depending on the animal species used in the
test. No differences for sex have been observed for absorption in either animals or humans.
However, human biomonitoring data do show that boys excrete significantly more uranium
than girls. In animal studies neonates took up more uranium than adults or older children.
Nutritional status, and in particular the iron content of the diet, have a marked influence on
absorption, and higher uranium levels in food intake also appear to increase the
absorption rate. If the pointers to an absorption mechanism competing with iron are
correct, these mechanisms could also explain the relatively high concentration and
chemical toxicity of uranium in the kidneys. It is here (and in the duodenum) that divalent
metal transporter 1 (DMT1), which is primarily responsible for the passage of iron (or
uranium?) through the cell membranes, is most strongly expressed. Quelle:

Inhalt englisch:
The concentration-dependent absorption behaviour of uranium was investigated with
surviving intestinal segments of rat jejunums, using an ex-vivo model. The results showed
a monotonic slightly nonlinear increase in absorption as uranium concentrations
increased. This trend was observed over the entire concentration range tested. In the lower
concentration range a slower linear ascent was observed while a steeper linear ascent was
found for the higher concentration range. Statistical fit was only slightly poorer for an
exponential function in the range of lower values and a logarithmic function in the range of
higher values. The proportion of uranium absorbed expressed as percent of uranium
concentrations in the perfusion solutions followed a monotonically increasing trend from
20 to around 200 ìg/l uranium in the perfusion solutions, which thereafter appears to
reach a plateau, as further increase towards concentrations around 400 ìg/l is not
substantial. The uranium concentration administered had no effect on the vitality and
consequently the functionality of the intestinal segments, measured in terms of active
glucose transport. The results imply that uranium concentrations of more than 20 ìg/l in
drinking water, for example, could lead to elevated absorption rates and thus to higher
internal exposures to consider when setting of Guideline values in this concentration
range. Quelle: http://www.sciencedirect.com

Inhalt englisch:
Common voles (Microtus arvalis) are common small mammals in some European
landscapes. They can be a major rodent pest in European agriculture and they are also a
representative generic focal small herbivorous mammal species used in risk assessment
for plant protection products. In this paper, common vole population dynamics, habitat and
food preferences, pest potential and use of the common vole as a model small wild
mammal species in the risk assessment process are reviewed. Common voles are a
component of agroecosystems in many parts of Europe, inhabiting agricultural areas
(secondary habitats) when the carrying capacity of primary grassland habitats is exceeded.
Colonisation of secondary habitats occurs during multiannual outbreaks, when population
sizes can exceed 1000 individuals ha-1. In such cases, in-crop common vole population
control management has been practised to avoid significant crop damage. The species'
status as a crop pest, high fecundity, resilience to disturbance and intermittent
colonisation of crop habitats are important characteristics that should be reflected in risk
assessment. Based on the information provided in the scientific literature, it seems
justified to modify elements of the current risk assessment scheme for plant protection
products, including the use of realistic food intake rates, reduced assessment factors or
the use of alternative focal rodent species in particular European regions. Some of these
adjustments are already being applied in some EU member states. Therefore, it seems
reasonable consistently to apply such pragmatic and realistic approaches in risk
assessments for plant protection products across the EU. © 2013 Society of Chemical

Inhalt englisch:
During the past two decades scientists, regulatory agencies and the European Commission
have acknowledged pharmaceuticals to be an emerging environmental problem. In
parallel, a regulatory framework for environmental risk assessment (ERA) of
pharmaceutical products has been developed. Since the regulatory guidelines came into
force the German Federal Agency (UBA) has been evaluating ERAs for human and veterinary
pharmaceutical products before they are marketed. The results show that approximately
10% of pharmaceutical products are of note regarding their potential environmental risk.
For human medicinal products, hormones, antibiotics, analgesics, antidepressants and
antineoplastics indicated an environmental risk. For veterinary products, hormones,
antibiotics and parasiticides were most often discussed as being environmentally relevant.
These results are in good correlation with the results within the open scientific literature of
prioritization approaches for pharmaceuticals in the environment. UBA results revealed
that prospective approaches, such as ERA of pharmaceuticals, play an important role in
minimizing problems caused by pharmaceuticals in the environment. However, the
regulatory ERA framework could be improved by (i) inclusion of the environment in the risk-
benefit analysis for human pharmaceuticals, (ii) improvement of risk management options,
(iii) generation of data on existing pharmaceuticals, and (iv) improving the availability of
ERA data. In addition, more general and integrative steps of regulation, legislation and
research have been developed and are presented in this article. In order to minimize the
quantity of pharmaceuticals in the environment these should aim to (i) improve the existing
legislation for pharmaceuticals, (ii) prioritize pharmaceuticals in the environment and (iii)
improve the availability and collection of pharmaceutical data.

Inhalt englisch:
Hows life? The answer can depend on the region in which you live. Many factors that
influence peoples well-being are local issues, such as employment, access to health
services, pollution and security. Policies that take into account regional differences beyond
national averages can therefore have a greater impact on improving well-being for the
country as a whole.
This report presents the OECD analytical framework for measuring well-being at the
regional level, as well as internationally comparable indicators on 9 well-being dimensions
for 362 regions across 34 OECD countries. It also sets out guidance for all levels of
government in using well-being measures to better target policies at the specific needs of
different communities. Drawing on a variety of practical experiences from OECD regions
and cities, the report discusses methodological and political solutions for selecting
regional well-being outcome indicators, monitoring the progress of regional well-being
performance over time, and implementing a process of multi-stakeholder engagement to
promote social change.
