ZUSAMMENFASSUNG
Die vorliegende Auswertung hatte zur Aufgabe, regulationsrelevante Literatur zur humantoxikologischen Bewertung von kurzkettigen Alkylphenolen und NSO-Heterozyklen zusammenzustellen und auf dieser Basis die Ableitbarkeit von humantoxikologisch begründete Geringfügigkeitsschwellen (GFS) für 10 Verbindungen einzuordnen. Die Ergebnisse sind in folgender Tabelle zusammengefasst.

1 HINTERGRUND UND AUFGABENSTELLUNG
Im Rahmen des Länderfinanzierungsprogramms Wasser, Boden und Abfall (unter LABO) wurden bereits drei Studien (Module 1 – 3, Projekt-Nr. B 2.09, B 2.10, B 2.11 und B 2.11a) durchgeführt, welche eine human- und ökotoxikologische Bewertung von kurzkettigen Alkylphenolen (SCAP) und NSO-Heterocyclen (NSO-HET) beinhalteten.
Für die humantoxikologische Bewertung wurde in diesen Vorprojekten zunächst nur eine eingeschränkte Literaturbasis (Endpunkte Kanzerogenität und Mutagenität) herangezogen, nämlich vorwiegend Daten aus Blotevogel et al. (2008) und aus einem ARCADIS-Abschlussbericht zum Vorhaben „Heterocyclische Aromaten und andere teeröltypische Schadstoffe im Grundwasser - TP 1“ (Blotevogel et al., 2007). Die dort gewählte Recherchestrategie ist nicht erläutert.
Aus diesem Grund wurde FoBiG, ebenfalls im Rahmen des Länderfinanzierungsprogramms „Wasser, Boden und Abfall“, beauftragt, für die folgenden 10 Stoffe eine umfassende Recherche der humantoxikologisch relevanten Daten durchzuführen. Es zeigte sich, dass zusätzlich zu den o.g. Quellen weitere Literatur gefunden werden konnte, allerdings in den meisten Fällen nicht ausreichend für eine Bewertung der Einzelsubstanz mit substanzspezifischen Daten ohne zusätzliche Struktur- und Analogiebetrachtungen.

2 METHODIK
2.1 LITERATURRECHERCHE UND DOKUMENTATION NACH „STANDARD EVALUATION PROCEDURE“ (SEP)
Zunächst wurde auf eine Registrierung unter REACH geprüft (ECHA, 2016).
Im nächsten Schritt wurde für alle 10 Stoffe wurde eine Literaturrecherche nach der FoBiG-internen SEP (siehe Anhang I: Datenrecherche nach SEP) durchgeführt. Strategie und Protokolle dieser Recherchen finden sich im Anhang 4 (Datenrecherche nach SEP). Allgemein wurde dabei zunächst mit der CAS-Nummer gesucht, bei unzureichenden Treffern zusätzlich mit dem/den Substanznamen. Eine Einengung wegen zu vieler unspezifischer Treffer war nur selten nötig, z.B. bei Indan, wo in der PubMed-Datenbank die MESH-Terms zur Selektion herangezogen wurden. Nur für Hydroxybiphenyl und Inden konnte eine ausreichende Bewertungsgrundlage festgestellt werden. Für die restlichen Stoffe wurde zusätzlich kostenpflichtig in den Datenbanken DIMDI und STN recherchiert. Diese Recherchen lieferten keine zusätzlichen Informationen, die für eine Bewertung herangezogen werden könnten.
2.2 IDENTIFIKATION ÄHNLICHER VERBINDUNGEN UND LITERATURRECHERCHE
Im Falle ungenügender Daten zu einem der 10 Stoffe sollten Daten zu ähnlichen Verbindungen identifiziert werden, um einen Bewertungsansatz basierend auf „Read-across“ zu prüfen bzw. zu ermöglichen. Je nach Stoff erfolgte dies nach unterschiedlichen Ansätzen:
a) Suche nach ähnlichen Verbindungen über eine Struktursuche in ChemIDplus (NLM, 2016b) – zunächst mit 80 % Ähnlichkeit, je nach Ergebnis auch mit 70 %. Allerdings sind die Suchkriterien in ChemIDplus nicht eindeutig definiert und es zeigte sich, dass damit z.B. bei den Dimethylchinolinen und den Dimethylthiophenen nicht alle relevanten Isomere erfasst werden. In diesem Fall wurden weitere, plausibel erscheinende zusätzliche Substanzen ergänzt.
b) Verwendung einer übergreifenden chemischen Bezeichnung für die Substanzgruppe oder eines übergeordneten Trivialnamens – Beispiel: um Daten zu ähnlichen Verbindungen für 2,4-Dimethylchinolin zu finden, wurde nach „methylquinoline“ und „Methychinolin“ bzw. „dimethylquinoline“ und „Dimethylchinolin“gesucht.
Nach Ansatz a) wurden aus dem Ergebnis der Struktursuche in ChemIDplus nach chemisch-toxikologischem Sachverstand basierend auf der Struktur besonders vielversprechende ähnliche Verbindungen ausgewählt und ggf. durch weitere plausibel erscheinende strukturverwandte Substanzen ergänzt (üblicherweise maximal 5). Die Suche folgte hier nicht streng der der Suchstrategie nach SEP. Im Falle einer vielversprechenden Verbindung (nennenswerte Anzahl relevanter Treffer in einer wichtigen Datenbank, z.B. PubMed) wurde die Recherche so lange weiterverfolgt (in weiteren Datenbanken bzw. freier Internetrecherche), bis ausreichend Literatur identifiziert wurde. Entsprechend wurden Verbindungen, bei denen ungenügende Treffer in einer oder zwei wichtigen Datenbanken resultierten, wurden nicht weiterverfolgt und für einen „Read-across“-Ansatz verworfen.
Nach Ansatz b) resultieren direkt Treffer in bibliographischen Datenbanken bzw. Google Scholar, deren Relevanz geprüft wurde. Strategie, Suchprotokolle und kurze Anmerkungen

zum Ergebnis finden sich im Anhang 4. Die Suche zu den verwandten Substanzen folgte hier nicht streng der Suchstrategie nach SEP. Ein vielversprechender Ansatz (nennenswerte Anzahl Treffer bei genügender Spezifität) wurde so lange weiterverfolgt (in weiteren Datenbanken bzw. freier Internetrecherche), bis ausreichend Literatur identifiziert wurde. Ansätze, bei denen ungenügende Treffer in einer oder zwei wichtigen Datenbanken resultieren, wurden nicht weiterverfolgt und verworfen.
2.3 ORIENTIERENDE BEWERTUNG NACH DEM THRESHOLD OF TOXICOLOGICAL CONCERN-KONZEPT (TTC)
Bei Substanzen mit schlechter Datenlage, bei denen auch mit einem „Read-across“-Ansatz keine Geringfügigkeitsschwelle ableitbar war, wurde die Ableitung einer tolerierbaren Körperdosis nach dem TTC-Konzept geprüft (ausgeführt mit Toxtree v.2.6.131 oder der OECD-Toolbox v.3.4.0 (OECD, 2016). Das TTC-Konzept wurde ursprünglich für Verpackungsmaterialien von Nahrungsmitteln entwickelt, findet aber mittlerweile auch auf Chemikalien in Nahrung, Kosmetika und Verbraucherprodukten Anwendung (siehe z.B. Barlow, 2005; Blackburn et al., 2005; EFSA, 2012; Kroes et al., 2005; Munro et al., 2008; SCCS/SCHER/SCENIHR, 2012). Für die unterschiedlichen Toxizitätsklassen (Cramer-Klassen) I, II und III wurden TTC von 1800, 540 und 90 μg/d abgeleitet, für gentoxische Substanzen ein TTC von 0,15 µg/d. Diesen Körperdosen entsprechen unter der Annahme von 2 L Trinkwasseraufnahme/d Trinkwasserkonzentrationen 900, 270, 45 und 75 ng/L. Bei fehlenden Daten zur Gentoxizität wurden nach dem TTC-Konzept die verschiedenen Kriterien eines Strukturverdachts für die Substanz und ergänzend auch für die strukturverwandten Verbindungen abgeprüft:

Bei positivem Strukturverdacht auf Gentoxizität und fehlenden Hinweisen auf sehr starke Gentoxizität könnte alternativ zum TTC-Werts für gentoxische Substanzen in Höhe von 75 ng/Liter das untere Abschneidekriterium der LAWA (LAWA, 2004), herangezogen werden, d.h. ein „Gesundheitlicher Orientierungswert“ (GOW) von 0,1 µg/Liter (100 ng/L) nach Dieter (2011).

The mutagenic activity of the polycyclic aromatic hydrocarbon-
containing fraction of several soot samples was measured in S.
typhimurium, using resistance to the purine analog 8-azaguanine as a
genetic marker. A postmitochondrial supernatant derived from livers of
phenobarbital- and/or Aroclor-pretreated male Sprague-Dawley rats was
incorporated into all assays to allow metabolism of promutagens to their
active forms. The mutagenic activity of the soot extracts ranged from 10-
20 times higher than could be accounted for by the amount of
benzo(a)pyrene present. The possibility that synergism occurs between
benzo(a)pyrene and some component in the soot extracts was
discounted by the finding of a simple additive relationship of
mutagenicity of a soot extract and added benzo(a)pyrene. To examine
the alternative explanation that other components of soot may have
undiscovered mutagenic activity, 70 polycylic aromatic hydrocarbons
were quantitatively assayed for their mutagenic potential; 34 of these
compounds induced a significant increase in the mutant fraction
resistant to 8-azaguanine. Of particular interest are the extreme
mutagenicities of perylene, cyclopenta(cd)pyrene and fluoranthene, all
of which exhibit greater mutagenicity than does benzo(a)pyrene at
equimolar concentrations. Using the measured activities of each
polycyclic aromatic hydrocarbon constituent in a kerosene soot, it is
possible to account for the mutagenic activity of the whole polycyclic
aromatic hydrocarbon fraction in terms of the additive mutagenicity of
its individual components.

There is evidence for the carcinogenicity of 4-methylquinoline, with
the development of liver tumors in male mice exposed as newborns
via three intraperitoneal injections and the initiation of skin tumors in
two studies in female mice. Further evidence of carcinogenic
potential is provided by clear evidence of mutagenicity in short-term
tests, induction of unscheduled DNA synthesis in rat hepatocytes in
vitro, and by strong chemical structural analogy with a known
carcinogen.

The mutagenicity of 33 quinolines in the Salmonella test using TA98 and
TA100 cells has been reported. Significant activity was found only with
TA100 cells. Quantitative structure-activity relationships (QSAR) could be
formulated using molecular orbital parameters or Hammett constants
and hydrophobic parameters for those compounds with substituents in
the 6, 7 and 8 positions. The QSAR points to the 2-position on the
quinoline ring as being the site for activation by S9 oxidation.

In Ames test, the quinoline derivatives, 6-nitroquinoline (6-NQ), 8-
nitroquinoline (8-NQ), 6-methylquinoline (6-NQ), 8-methylquinoline (8-
MQ) and 8-hydroxyquinoline (8-HQ) are mutagenic, whereas 5,7-
dibromo-quinoline (5,7-DQ) is not. These compounds were administered
to groups of male and female F344 rats in their diet for 104 weeks. 8-NQ
induced forestomach tumors in male and female rats: it induced
squamous cell papillomas in 28 (93.3%) of 30 males and in 36 (97.3%) of

37 females, and squamous cell carcinomas in 20 (66.7%) of 30 males and
24 (64.9%) of 37 females. Thus there was no sex difference in its
carcinogenicity. 6-NQ induced only focal hyperplasia of the forestomach
at an incidence of 30.0% in male rats and 52.8% in female rats. These
results indicate a discrepancy between the mutagenicities of quinoline
derivatives and their carcinogenicities to rats: only 8-NQ was
carcinogenic to F344 rats.

Quinoline in the presence of microsomal activation exhibits mutagenic
activity in Salmonella typhimurium TA100 and induces unscheduled DNA
synthesis (UDS) in rat hepatocytes. Structure-activity studies were
performed to determine those positions on quinoline critically
associated with its genotoxicity. In assays performed to determine the
ability of 2-, 4-, 6- and 8-methylquinoline to induce UDS, only 4- and 8-
methylquinoline produced a positive response. This represents an
improved correlation for these methylquinolines with tumorigenic
activity as compared to that previously observed with mutagenic activity
in S.typhimurium TA100. …. These data indicate that positions 1 - 3 in
quinolone are critical sites associated with its genotoxicity.

Quinoline and all 7 positional isomers of methylquinoline were assayed
for tumor-initiating activity on the skin of SENCAR female mice with
promotion by tetradecanoyl phorbol acetate. The total initiation dose of
either quinoline or the isomeric methylquinolines was 7.5 mg per mouse.
Quinoline induced tumors in 53% of the mice (0.73 tumors per animal).
While 2-, 3-, 5-, 6- and 7-methylquinoline did not exhibit significant
tumorigenic activity in this assay, 4-methylquinoline induced tumors in
45% of the mice (0.90 tumors per animal). 8-Methylquinoline induced
tumors in 45% of the mice (0.66 tumors per animal).

4-Methychinolin ist stark gentoxisch
4-Methylquinoline (4-MeQ) showed an extraordinarily potent
mutagenicity when compared to quinoline and isomeric
methylquinolines. The major metabolite of 4-MeQ was 4-
hydroxymethylquinoline, which was not mutagenic under the assay
condition employed. Deuteration of the methyl group of 4-MeQ resulted
in a decrease in the amount of the hydroxymethyl metabolite and an
increase in mutagenicity, indicating that hydroxylation of the substituent
methyl group is a detoxication process. A 3-chloro derivative of 4-MeQ
was proven to be non-mutagenic. 4-Ethyl-quinoline, as well as 4-
hydroxymethylquinoline, was much less mutagenic than 4-MeQ. Taking
account of the structure-mutagenicity relationship, a possible
mechanism is proposed for the potent mutagenic potential of 4-MeQ.

Metabolismus von 8-Methychinolin
The hepatic microsomal metabolism of the carcinogenic 8-

methylquinoline (8MQ) and its noncarcinogenic isomer, 6-
methylquinoline (6MQ), were compared for preparations from control
rats and rats pretreated with phenobarbital or 3-methylcholanthrene.
For each compound the alcohol was the major metabolite, constituting
50-75% of 6MQ metabolites and 60-85% of 8MQ metabolites. Three
phenols for 6MQ and two for 8MQ were seen. The latter formed two
dihydrodiols which constituted 1-5% of metabolites while dihydrodiol
products were not identified for the nontumorigenic 6MQ. Epoxides
were observed for 6MQ while 8MQ afforded an N-oxide
(Sengoku et al., 1989)
Mechanistische Betrachtungen zur metabolischen Aktivierung
3-Fluoro, 2-chloro and 3-chloro derivatives of the potently mutagenic
quinoline were deficient in mutagenicity in Salmonella typhimurium
TA100, whereas all the other fluoro and chloro derivatives examined,
including 4-chloroquinoline, were mutagenic. These results suggest that
the ultimate structure of genotoxic quinoline may be the 2,3-dihydro-
2,3-epoxide of the 1,4-hydrate form of quinoline. This is supported by
the finding that, among methyl-substituted quinolines, the mutagenicity
of the 2-methyl isomer was weakest and that of the 4-methyl isomer
highest. The present study provides support for the view that a fluorine
substitution at the site of the oxidation leading to the genotoxic
metabolite may deprive the aromatic hydrocarbons of their genotoxicity
without affecting other biological properties of the parent hydrocarbons.

3.1.6 STRUKTUR-WIRKUNGSBEZIEHUNGEN
Zur Überprüfung der Struktur-Wirkungsbeziehungen wurde mittels der der Toxtree-Software2 die Toxizitätsklassen nach Cramer für Chinolin sowie die oben angeführten Dimethylchinoline und Ethylmethylchinolin abgeprüft. Für sämtliche Verbindungen ergab sich die Cramer-Klasse II (TTC: 900 µg/d, 450 µg/L). Weiterhin wurde mittels der ebenfalls in Toxtree enthaltenen „Carcinogenicity and genotoxicity rulebase by ISS“, der „In vitro mutagenicity alerts by ISS“ und der „Structure Alerts for the in vivo micronucleus assay in rodents“ die Ähnlichkeit der verwandten Substanzen zu 2,4-Dimethylquinolin geprüft. Zwar ergaben sich keine positiven Hinweise (Alerts) für alle abgeprüften Strukturmerkmale auf mögliche Gentoxizität oder Kanzerogenität (was den experimentellen Beobachtungen widerspricht), jedoch zeigt diese Auswertung, dass alle Substanzen hinsichtlich ihrer Strukturmerkmale gleich beurteilt werden. Zur Absicherung wurde dies auch noch mit der

QSAR Toolbox v.3.4.0 (OECD, 2016) überprüft, hier ergaben sich ebenfalls keine positiven strukturellen Alerts für „DNA alerts for AMES by OASIS v.1.4“., „DNA alerts for CA and MNT by OASIS v.1.1“, „in vitro mutagenicity (Ames test) alerts“, „in vivo mutagenicity (Micronucleus) alerts by ISS“ und „Carcinogenicity and genotoxicity rulebase by ISS“. Ein positiver Alert zeigte sich für „DNA binding by OASIS v1.4 (direct acting epoxides formed after metabolic activation)“, nicht aber in „DNA binding by OECD“. Der Parameter DNA-Bindung ist nicht in toxtree enthalten. Der positive Alert (OASIS v1.4) steht in Einklang mit dem beobachteten Metabolismus (siehe im Folgenden).
Die niedrigere mutagene Aktivität von an der Position 2 methylsubstituierten Chinolinen geht möglicherweise darauf zurück, dass bei Chinolin eine Epoxidierung des Rings an den C-Atomen 2 und 3 zu gentoxischen Metaboliten führt (EPA, 2016), was durch die Methylierung in Position 2 zu sterischer Hinderung und damit weniger effizienter Aktivierung führen könnte.
3.1.7 SCHLUSSFOLGERUNG
Die Datenlage zu 2,4-Dimethylchinolin ist ungenügend für eine stoffspezifische Bewertung.
Es liegt ein Ames-Test mit einem negativen Ergebnis vor. Zu monomethylierten Isomeren konnten mehrere potentiell relevante Literaturstellen v.a. zur Kanzerogenität und Mutagenität gefunden werden. Die einfach methylierten Chinoline unterscheiden sich je nach Substitutionsposition in ihrer mutagenen Aktivität: nach NTP war die mutagene Aktivität des 4-Methylchinolin höher als bei Chinolin, andrerseits aber die des 2-Methylisomers geringer. Im Ames-Test waren 2,4-, 2,6- und 2,7 Dimethylchinolin nicht mutagen. Es ist unklar, ob bzw. wie sich Wirkungen der monomethylierten Spezies auf 2,4-Dimethylchinolin übertragen lassen. Offensichtlich unterschieden sich die methylierten Chinoline (hinsichtlich der mutagenen Wirkung) von den methylierten Benzolderivaten Toluol und Xylol, welche beide (im Gegensatz zu Benzol) keine gentoxischen und auch keine kanzerogenen Effekte zeigen (siehe z.B. EPA, 2016). Ein „Read-across“ würde angesichts der lückenhaften Datenlage ebenfalls keine Bewertung für das 2,4-Dimethylchinolin ermöglichen.
Bei dieser Sachlage ist eine Orientierung an der Toxizität von Chinolin zu diskutieren, d.h. der relevanten krebserzeugenden Wirkung, welche in allen vorliegenden Bewertungen als kritischer Endpunkt angesehen wird. Offensichtlich sind die nichtkanzerogenen Effekte weniger kritisch. Chinolin ist gut untersucht, es liegt eine lineare Krebsrisikoabschätzung der EPA vor, ein an dieser Risikoabschätzung orientierter Trinkwasserwert der LAWA und weiterhin im Registrierungsdossier ein DMEL (Derived Minimum Effect Level), d.h. ein risikobasierter Arbeitsplatzgrenzwert für kanzerogene Effekte auf Basis einer T25 (abgeschätzte Dosis, bei der 25 % Tumoren erwartet werden). Ob sich dieser nach vertiefter Betrachtung in ein Risiko bei oraler Exposition umrechnen lässt, muss anhand der verfügbaren Daten im Registrierungsdossier geprüft werden.
Die Risikoabschätzung der EPA in Höhe von 9 x 10-5 pro µg/L resultiert bei einem Risikoniveau von 10-6 in einer Trinkwasserkonzentration von 0,011 µg/L (11 ng/L), d.h. niedriger als der TTC für kanzerogene Effekte (75 ng/L). Angesichts der Unsicherheiten der Übertragung der Daten von Chinolin wäre deshalb auch denkbar, für die Bewertung von 2,4-Dimethylchinolin den TTC anzuwenden.

Zwar stellt die Annahme einer potenziellen krebserzeugenden Wirkung für 2,4-Dimethylchinolin eine konservative Vorgehensweise („worst-case“) dar, da es, im Gegensatz zu Chinolin, im Ames-Test eine negative Wirkung zeigte. Angesichts fehlender Daten zu weiteren gentoxischen Endpunkten (z.B. Mutagenität in Säugerzellen sowie Klastogenität in vitro und in vivo) scheint diese Vorgehensweise jedoch gerechtfertigt.
In die Bewertung nicht einbezogen werden kann der Endpunkt endokrine Wirkungen: sowohl Chinolin als auch 6-Methylchinolin zeigten im CALUX-Assay (Brinkmann et al., 2014) östrogene Aktivität, wobei 6-Methylchinolin eine etwas höhere Aktivität besaß als das Grundgerüst. 2,4-Dimethychinolin wurde nicht untersucht. Diese in vitro-Befunde ergeben Verdachtsmomente auf endokrine Wirkungen auch für 2,4-Dimethylchinolin, welche aber mangels geeigneter in vivo-Daten noch nicht in die Bewertung einfließen können.

Es konnte keine bewertungsrelevante Literatur identifiziert werden. Studien von Mosier et al. (2003) und Du et al. (2008) umfassten zwar experimentelle Daten zu 140 Thiophenderivaten im SOS-Chromo-Test, jedoch keine der hier relevanten Verbindungen.

About 40 coffee aroma constituents belonging to the classes of
dicarbonyls, sulphur-containing compounds, furfuryls, N-heterocyclics
and others were systematically evaluated in three Ames tester strains.
Only aliphatic dicarbonyl compounds showed notable direct mutagenic
activity, which mainly affected 'base-pair substitution' in Ames tester
strains TA100 and TA102. Very weak effects were also seen with some N-
heterocyclics, mainly affecting frameshift tester strain TA98 upon
metabolic activation. However, it was shown that these N-heterocyclics
do not contribute substantially to the mutagenicity in coffee. The
hydrogen peroxide and methylglyoxal contents of coffee were
determined up to 26 hr after preparation. Their concentrations tended
to decrease whereas mutagenic activity decreased significantly with time
in tester strains TA100 and TA102. It is concluded that several highly
labile coffee constituents contribute to the bacterial mutagenicity and

also that the synergism between hydrogen peroxide and methylglyoxal is
not the main factor. The absence of coffee mutagenicity/carcinogenicity
in rodents with these highly reactive coffee aroma compounds can be
explained in part by detoxification of microsomal enzyme systems.

Für die Bewertung der Toxizität nach wiederholter Exposition wurde im OECD-Dokument ein NOAEL von 25 mg/kg x d für Lebertoxizität in Ratten angegeben, diese Dosis stellt aber den LOAEL für Fertilitätseffekte in weiblichen Ratten dar (Studie nach OECD Guideline 422: Combined Repeated Dose Toxicity Study with the Reproduction / Developmental Toxicity Screening Test). In vitro Gentoxizitätstests (Ames-Test, Chromosomenaberrationstest in Hamsterlungenzellen) erbrachten negative Resultate, im Mauslymphomtest war Thiophen zwar mutagen, aber nur bei Konzentrationen, welche massive Zytotoxizität verursachten. OECD beurteilte die Substanz als nicht gentoxisch. In vivo-Tests und Kanzerogenitätsstudien liegen nicht vor.
3.2.6 STRUKTUR-WIRKUNGSBEZIEHUNGEN
Zur Überprüfung der Struktur-Wirkungsbeziehungen wurde mittels der der Toxtree-Software4 die Toxizitätsklassen nach Cramer für Thiophen sowie die oben angeführten Dimethylthiophene abgeprüft. Für sämtliche Verbindungen ergab sich die Cramer-Klasse III (TTC 90 µg/d, 45 µg/L). Diese Einstufung weicht von der in EFSA (2015, s.o.) berichteten ab, wo für 2,5-Dimethylthiophen, 2- und 3-Methythiophen eine Einordnung in Klasse II (540 mg/d, 270 mg/L) refereiert ist. Die Hintergründe für diese Abweichung liegen möglichweise darin begründet, dass sich die EFSA methodisch auf eine Veröffentlich von 1978 bezieht, also möglicherweise nicht auf der aktuellen Toxtree-Version. Weiterhin wurde mittels der ebenfalls in Toxtree enthaltenen „Carcinogenicity and genotoxicity rulebase by ISS“, der „In vitro mutagenicity alerts by ISS“ und der „Structure Alerts for the in vivo micronucleus assay in rodents“ die Ähnlichkeit der verwandten Substanzen zu 2,3-Dimethylthiophen geprüft. Zwar ergaben sich keine positiven Hinweise (Alerts) für alle abgeprüften Strukturmerkmale auf mögliche Gentoxizität oder Kanzerogenität (was den experimentellen Beobachtungen entspricht). Diese Auswertung ergab, dass alle Substanzen hinsichtlich ihrer Strukturmerkmale gleich beurteilt werden. Zur Absicherung wurde dies auch noch mit der QSAR Toolbox v.3.4.0 (OECD, 2016) überprüft, hier ergaben sich ebenfalls keine positiven strukturellen Alerts für „DNA alerts for AMES by OASIS v.1.4“., „DNA alerts for CA and MNT by OASIS v.1.1“, „in vitro mutagenicity (Ames test) alerts“, „in vivo mutagenicity (Micronucleus) alerts by ISS“, „Carcinogenicity and genotoxicity rulebase by ISS“ und „DNA binding by OASIS v1.4 (direct acting epoxides formed after metabolic activation)“. Für „DNA binding by OECD“ (Michael Addition nach metabolischer Aktivierung) waren die Ergebnisse unterschiedlich: 2,5-Dimethylthiophen erbrachte einen negativen Alert, die 2,3- und 2,4-Isomere sowie 2- und 3-Methylthiophen und Thiophen hatten einen positiven Alert. Damit ergibt sich für das 2,3-Isomer und auch Thiophen ein Verdacht auf potenzielle Gentoxizität und Kanzerogenität. Dieser steht aber, im Gegensatz zu Chinolin (siehe 0), im Widerspruch zu Ames-Tests mit negativem Ergebnis. Auch die Gentoxizitätstests zu Thiophen stehen in Widerspruch zu dem strukturellen Verdachtsmoment. Insofern wird diesem Alert keine

entscheidende Bedeutung zugemessen und 2,3-Methylthiophen als vermutlich nicht gentoxisch bewertet.
3.2.7 SCHLUSSFOLGERUNG
Die Datenlage zu 2,3-Dimethylthiophen ist ungenügend für eine stoffspezifische Bewertung.
Zu 2,3-Dimethylthiophen konnten nur Daten zur akuten oralen Toxizität und Reizwirkung gefunden werden. 2- und 3-Methyltiophen waren in Ames-Tests nicht mutagen. Thiophen wird nach OECD-SIDS nicht als gentoxisch angesehen, Kanzerogenitätsstudien liegen nicht vor. Damit sollte in erster Betrachtung die Gentoxizität und Kanzerogenität für die Bewertung der methylierten Thiophene keine Rolle spielen. Nachdem sowohl Thiophen als auch 2,3-Dimethylthiophen nach der Toxtree-Software5 in Cramer-Klasse III (TTC 90 µg/d, 45 µg/L) eingeordnet werden, was auf eine ähnlich hohe Toxizität hinweist, wird angesichts mangelnder stoffspezifischer Daten hilfsweise eine Übernahme eines Richtwertes von Thiophen vorgeschlagen. Zur Überprüfung der Struktur-Wirkungsbeziehungen wurde mittels der der Toxtree-Software6 die Toxizitätsklassen nach Cramer für Thiophen sowie die oben angeführten Dimethylthiophene abgeprüft. Für sämtliche Verbindungen ergab sich die Cramer-Klasse III (TTC 90 µg/d, 45 µg/L). Weiterhin wurde mittels der ebenfalls in Toxtree enthaltenen „Carcinogenicity and genotoxicity rulebase by ISS“, der „In vitro mutagenicity alerts by ISS“ und der „Structure Alerts for the in vivo micronucleus assay in rodents“ die Ähnlichkeit der verwandten Substanzen zu 2,5-Dimethylthiophen geprüft. Zwar ergaben sich keine positiven Hinweise (Alerts) für alle abgeprüften Strukturmerkmale auf mögliche Gentoxizität oder Kanzerogenität (was den experimentellen Beobachtungen entspricht). Diese Auswertung ergab, dass alle Substanzen hinsichtlich ihrer Strukturmerkmale gleich beurteilt werden. Zur Absicherung wurde dies auch noch mit der QSAR Toolbox v.3.4.0 (OECD, 2016) überprüft, hier ergaben sich ebenfalls keine positiven strukturellen Alerts für „DNA alerts for AMES by OASIS v.1.4“., „DNA alerts for CA and MNT by OASIS v.1.1“, „in vitro mutagenicity (Ames test) alerts“, „in vivo mutagenicity (Micronucleus) alerts by ISS“, „Carcinogenicity and genotoxicity rulebase by ISS“ und „DNA binding by OASIS v1.4 (direct acting epoxides formed after metabolic activation)“. Für „DNA binding by OECD“ (Michael Addition nach metabolischer Aktivierung) waren die Ergebnisse unterschiedlich: 2,5-Dimethylthiophen erbrachte einen negativen Alert, die 2,3- und 2,4-Isomere sowie 2- und 3-Methylthiophen und Thiophen hatten einen positiven Alert. 2,5-Dimethylthiophen wird damit als vermutlich nicht gentoxisch bewertet.

The systemic and neurobehavioral effects of benzo(b)thiophene
(routinely referred to as benzothiophene) were studied in rats following
13-wk oral exposure. Male and female Sprague-Dawley rats (10 animals
per group) were fed diet containing 0.5, 5, 50, or 500 ppm
benzothiophene for 13 wk. Control animals were given rat feed plus
vehicle (corn oil) only. No clinical signs of toxicity and neurobehavioral
effects were observed using screening tests that included cage-side
observations, righting reflex, open field activities, and forelimb and
hindlimb grip strength. Elevated serum aspartate aminotransferase
activity and bilirubin level were observed in highest dose females. Except
for a statistically significant decrease in hematocrit in the highest dose
males, benzothiophene exerted no marked effects on hematological
parameters. Benzothiophene exposure did not result in alterations in
hepatic alkaline phosphatase activity, or the typical hepatic phase I
(aniline hydroxylase, ethoxyresorufin O -deethylase, pentoxyresorufin O
-dealkylase, aminopyrine N -demethylase) and phase II
(UDPglucuronosyltransferase, glutathione S -transferase) drug-
metabolizing enzyme activities. No significant elevation in urinary
ascorbic acid, protein, and N -acetylglucosaminidase activity was
detected in the treated animals. Peribiliary fibrosis was the most
significant histological change and occurred in the liver of females in the
50 and 500 ppm groups. Mild epithelial hyperplasia in the renal pelvis
was detected in the majority of 5 and 50 ppm females, with epithelial
hyperplasia in the urinary bladder observed in the 50 ppm females. In
males, increased incidence and severity of mild binucleation of
hepatocytes and mild thickening of the basement membrane in kidney
cortex were observed at 500 ppm. Benzothiophene was not detected in
the urine of high-dose animals at the termination of the experiment.
Based on the kidney, hepatic, and hematocrit changes, the no-observed-
adverse-effect level (NOAEL) in the diet was determined to be 0.5 ppm
(0.04 mg/kg/ d) for females and 50 ppm (3.51 mg/kg/ d) for males.

Dieses „Read-across“ scheint gerechtfertigt, da sowohl für 3-Methylbenzothiophen als auch Benzothiophen die gleiche Cramer-Klasse III (TTC 90 µg/d, 45 µg/L), also in erster Näherung eine vergleichbare Toxizität vorausgesagt wird und die Struktur-Wirkungsanalyse auch keine strukturellen Unterschiede hinsichtlich möglicher gentoxischer oder kanzerogener Wirkungen ergab.

Groups of 15 male and 15 female Sprague-Dawley-derived OFA rats 4–5
weeks of age were administered 2,3-dimethylbenzofuran (purity not
given) at 0.6 mg/kg bw per day in olive oil by gavage 7 days per week for
a period of 13 weeks. A concurrently maintained control was
administered the vehicle substance. Water and food were provided ad

libitum. Animals were subject to daily examination and observations of
behaviour. Body weights for each rat and food consumption for each
cage of four animals were measured weekly. Haematology and serum
biochemistry were performed for eight males and eight females at weeks
4 (serum biochemistry limited to blood urea nitrogen) and 13. At 13
weeks, 16 male and 16 female control and treated rats were necropsied.
The liver, kidneys, spleen, heart, adrenal glands, testes and ovaries were
weighed. Major tissue types were preserved for histopathological
examination. Clinical examinations revealed no differences in the
mortality, behaviour, body weight gain or food consumption for either
test group in comparison with the corresponding group of control
animals. Administration of 2,3-dimethylbenzofuran for 13 weeks showed
no effect on haematology tests. Serum biochemistry revealed a slight
increase in ALP activity in males and a slight increase in bilirubin levels in
males and females administered the substance. A decrease in the serum
glucose level of test group females was also observed. At necropsy,
pericarditis or an induration of the thymus was observed in three treated
and four control animals. In the opinion of the author, these findings
were likely a result of intubation error. All organ weights were
comparable between test and control animals, and histology revealed no
morphological changes that could be attributed to the administration of
2,3-dimethylbenzofuran.

Die Kanzerogenität von Benzofuran stellt den kritischen toxikologischen Endpunkt dar. Auf Basis der vorliegenden Daten ist in vertiefter Betrachtung eine quantitative Krebsrisikobewertung möglich.
3.6.5 STRUKTUR-WIRKUNGSBEZIEHUNGEN
Die Überprüfung der Struktur-Wirkungsbeziehungen wurde mittels der QSAR Toolbox v.3.4.0 (OECD, 2016) durchgeführt (die bei den Stoffbewertungen 3.1 bis 3.3 zusätzlich verwendete

Bewertung mittels Toxtree wurde nicht durchgeführt, da für die dort erfassten Parameter identische Ergebnisse erhalten wurden). Es ergaben sich für die einfach und dimethylierten Benzofurane und auch für Benzofuran durchgängig die Cramer-Klasse III (90 µg/d, 45 µg/L) sowie keine positiven strukturellen Alerts für „DNA alerts for AMES by OASIS v.1.4“., „DNA alerts for CA and MNT by OASIS v.1.1“, „in vitro mutagenicity (Ames test) alerts“, „in vivo mutagenicity (Micronucleus) alerts by ISS“, „Carcinogenicity and genotoxicity rulebase by ISS“, „DNA binding by OASIS v1.4 (direct acting epoxides formed after metabolic activation)“ und „DNA binding by OECD“ (Michael Addition nach metabolischer Aktivierung). Dies steht in Gegensatz zu den experimentellen Beobachtungen zum gentoxischen und kanzerogenen Benzofuran. Dennoch ergab die Auswertung, dass alle betrachteten Substanzen hinsichtlich ihrer Strukturmerkmale gleich beurteilt werden.
3.6.6 SCHLUSSFOLGERUNG
Die Datenlage zu 3-Methylbenzofuran ist ungenügend für eine stoffspezifische Bewertung.
Eine subchronische Studie zum 2,3-Dimethylbenzofuran mit einem NOAEL von 0,6 mg/kg x d genügt mit nur einer getesteten Dosis nicht heutigen Standards zur Ableitung eines Richtwertes.
Die Struktur-Wirkungsanalyse ergab für 3-Methylbenzofuran, strukturverwandte Verbindungen und Benzofuran in erster Näherung eine vergleichbare Toxizität (Cramer-Klasse III, 90 µg/d bzw. 45 µg/L). Hinsichtlich der Gentoxizität differierten die Aussagen der Struktur-Wirkungsanalyse und die experimentellen Befunde: keine Hinweise auf einen potenziell gentoxischen oder kanzerogenen Wirkungsmechanismus stehen im Gegensatz zu den experimentellen Befunden zu Benzofuran, die gentoxische und kanzerogene Effekte belegen. Insofern kann angenommen werden, dass auch die methylierten Derivate ähnliche Eigenschaften besitzen. Bei dieser Sachlage kann bei fehlenden stoffspezifischen Daten hilfsweise eine Bewertung auf Basis der quantitativen Krebsrisikoabschätzung zu Benzofuran durchgeführt werden. Die Qualität dieser Krebsrisikoabschätzung wäre in einer vertieften Betrachtung zu überprüfen, da sie auch Lebertumoren in B6C3F1-Mäusen einbezieht, welche generell als besonders sensitiv für die Entwicklung von Lebertumoren angesehen werden. Nach ATSDR erfolgt die metabolische Aktivierung über eine Epoxidierung der Doppelbindung des Furanringes. Es liegt die Vermutung nahe, dass die Methylierung der Doppelbindung bei 3-Methylbenzofuran zu einer sterischen Hinderung der Epoxidierung und damit einer geringeren oder verlangsamten metabolischen Aktivierung zu gentoxischen oder kanzerogenen Metaboliten führt. Insofern kann vermutet werden, dass die Übernahme der Risikoschätzung von Benzofuran zu keiner Risikounterschätzung führt. Der konservative Charakter dieses „Read-across“-Ansatzes kann nicht abgeschätzt werden. Aus der Risikoabschätzung der OEHHA ergäbe sich für ein Risiko von 10-6 eine korrespondierende Trinkwasserkonzentration von 56 ng/L, was in erster Näherung identisch mit dem TTC für gentoxische Substanzen in Höhe von 75 ng/l ist.
Allerdings zeigen sich auch bei Betrachtung des Endpunkts endokrine Wirkungen die Grenzen dieses „Read-across“-Ansatzes: während Benzofuran selbst im CALUX-Assay (Brinkmann et al., 2014) keine östrogene Aktivität besaß, zeigten 2-Methylbenzofuran und in noch höherem Ausmaß 2,3-Dimethylbenzofuran östrogene Aktivität. Diese in vitro-Befunde ergeben Verdachtsmomente auf endokrine Wirkungen, welche aber mangels geeigneter in vivo-Daten noch nicht in die Bewertung einfließen können.

3.7.6 SCHLUSSFOLGERUNG
Die Datenlage zu 2-Methyldibenzofuran und auch der berücksichtigten anderen Methylisomere, Dimethylisomere und Dibenzofuran ist ungenügend für eine stoffspezifische Bewertung.
Die experimentellen Daten zu Dibenzofuran deuten darauf hin, dass die Substanz in vitro nicht mutagen wirkt, auch nicht bei metabolischer Aktivierung. Allerdings zeigt die Struktur-Wirkungsanalyse für 2-Methyldibenzofuran und die anderen betrachteten strukturverwandten Substanzen aufgrund der Zugehörigkeit zur Klasse der heterozyklischen

PAK positive Alerts für in vitro und in vivo Mutagenität sowie Kanzerogenität. Nachdem keine in vivo-Daten vorliegen, kann dies nicht entkräftet werden. Dies würde in einem TTC für gentoxische Substanzen von 0,15 µg/d (75 ng/l) resultieren.
Bei Blotevogel et al. (2007) wurde für die Methylbenzodifurane eine Geringfügigkeitsschwelle von 0,01 µg/L, der Untergrenze bei LAWA (2004), abgeleitet. Bei Blotevogel et al. wird dies allerdings damit begründet, dass das unsubstituierte Dibenzofuran als mutagen und gentoxisch bekannt ist und für Methyldibenzofuran eine ähnlich hohe humantoxikologische Aktivität anzunehmen sei. Die Aussagen zum Dibenzofuran stehen im Gegensatz zu den bei der Recherche gefundenen experimentellen Ergebnissen. Zudem gilt dieser Wert von 0,01 µg/L unter humantoxikologischen Aspekten definitionsgemäß für stark gentoxische Substanzen, was anhand der vorliegenden Daten nicht wahrscheinlich ist (ökotoxikologische Aspekte, welche ebenfalls zu einer Untergrenze von 0,01 µg/L führen, gehen in die vorliegende Bewertung nicht ein). Bei dieser Sachlage wird bei fehlenden stoffspezifischen Daten hilfsweise eine Bewertung auf Basis des TTC-Ansatzes für gentoxische Substanzen vorgeschlagen. Dies würde eine Geringfügigkeitsschwelle von 75 ng/L bedeuten (0,15 µg/Person und Tag unter Berücksichtigung von 2 L Trinkwasseraufnahme). Alternativ könnte unter humantoxikologischen Aspekten der etwas höhere „Gesundheitliche Orientierungswert“ (GOW) von 0,3 µg/Liter (LAWA, 2004) verwendet werden.

(Bomhard et al., 2002) Ortho-phenylphenol (OPP) and its sodium (SOPP) and potassium (POPP)
salts are used as fungicides and disinfectants. Due to the widespread use
of especially OPP and SOPP, the potential for consumer exposure and
some "critical" findings the toxicological database is quite extensive and
complex. In experimental animals toxicity after single oral and dermal

administration of these compounds is low. For the skin and mucous
membranes, OPP has to be considered as irritating, and SOPP and POPP
as corrosive. A large number of chronic toxicity and reproduction studies
did not show any indication of oestrogen-like or other endocrine effects
of OPP in the mammalian organism. No teratogenic effects were
observed after the administration of OPP or SOPP in rats, mice, and
rabbits. In two-generation studies in rats, OPP did not affect
reproduction. The available data do not suggest a relevant potential for
immunotoxic properties. The administration of high dietary
concentrations of OPP to mice up to 2 years induced hepatocellular
changes indicative of adaptations to metabolic demands, zonal
degeneration, focal hepatocellular necrosis, and/or pigmentation of the
liver. Only in male mice of one study, using a strain prone to develop
hepatocellular tumors at high spontaneous incidences, the incidence of
hepatocellular adenomas was increased. The incidence of hepatocellular
carcinomas was not affected by treatment. The urothel of the urinary
bladder (at very high doses also of the renal pelvis and the papilla) is the
main target tissue after the repeated oral exposure of rats. The changes
initially consist of increased mitosis, followed by simple epithelial
hyperplasia, developing to a papillary and/or nodular form, later on to
papillomas and transitional carcinomas. Crystals or stones in the bladder
do not play a decisive role in this cascade. SOPP is more effective than
OPP in this respect. Male rats are much more sensitive than females. In
mice, hamsters, guinea pigs, and dogs, urothelial lesions do not develop
even at very high oral dose levels. The findings in rats explain why there
is a large genotoxicity/mutagenicity data base not only for OPP and SOPP
but also for their metabolites on nearly all kinds of endpoints/targets.
The weight of evidence suggests that genotoxicity of OPP/SOPP or their
metabolites does not play a decisive role for the carcinogenicity at the
urothel. Among them are lack of DNA binding of OPP to the rat bladder
epithelium, the differences between OPP and SOPP, between male and
female rats, between rats and mice (despite roughly comparable
toxicokinetics), as well as the fact that tumors develop only at dose levels
inducing hyperplasias. In addition, the strong dependence of the
incidence and severity of the nonneoplastic and neoplastic bladder
changes on urinary pH values (modified by feeding of ammonium
chloride or sodium hydrogen carbonate) is consistent with the
hypothesis of a nongenotoxic mode of action. Finally, there is no
correlation between the urinary concentration of OPP or its metabolites
and the incidence of hyperplasias/tumors in the urinary bladder. Both
tumorigenic effects in rats and male mice are considered to represent
high-dose, sex- and/or species-specific phenomena, based on
nongenotoxic mechanisms of action and therefore allow the conclusion
that the conventional margin of safety approaches are appropriate when
assessing the risk of applications of OPP and its salts.

The Environmental Protection Agency (EPA or the Agency) has
completed the human health and environmental risk assessments for 2-
phenylphenol (orthophenylphenol or OPP) and its salts and is issuing its
risk management decision and tolerance reassessment. The risk
assessments, which are summarized below, are based on the review of
the required target database supporting the use patterns of currently
registered products and additional information received through the
public docket. After considering the risks identified in the revised risk
assessments, comments received, and mitigation suggestions from
interested parties, the Agency developed its risk management decision
for uses of OPP and salts that pose risks of concern. As a result of this
review, EPA has determined that OPP and salts-containing products are
eligible for reregistration, provided that risk mitigation measures are
adopted and labels are amended accordingly. That decision is discussed

3.9.3 VERWANDTE SUBSTANZEN
Aus ChemID (NLM, 2016b) wurden mit einer 80 % Übereinstimmung der Struktur folgende weitere Substanzen angezeigt:
Indan (alkane) mit identischer Struktur (CAS-Nr. 56573-11-6)
Bei einer Erweiterung auf 70 % Übereinstimmung ergab sich zusätzlich:
4-Methylindan (CAS-Nr. 824-22-6)
Weiterhin wurde die OECD-Toolbox hinsichtlich der in Serve et al. (1990) genannten Metaboliten (indan-5-ol, indan-1-ol; indan-2-one, indan-2-ol) ausgewertet.

3.9.4 STRUKTUR-WIRKUNGSBEZIEHUNGEN
Die Überprüfung der Struktur-Wirkungsbeziehungen wurde mittels der QSAR Toolbox v.3.4.0 (OECD, 2016) durchgeführt (die bei den Stoffbewertungen 3.1 bis 3.3 zusätzlich verwendete Bewertung mittels Toxtree wurde nicht durchgeführt, da für die dort erfassten Parameter identische Ergebnisse erhalten wurden). Es ergaben sich für Indan und seine Metaboliten (siehe 3.9.3) keine positiven strukturellen Alerts für „DNA alerts for AMES by OASIS v.1.4“., „DNA alerts for CA and MNT by OASIS v.1.1“, „in vitro mutagenicity (Ames test) alerts“, „in vivo mutagenicity (Micronucleus) alerts by ISS“, „Carcinogenicity and genotoxicity rulebase by ISS“, „DNA binding by OASIS v1.4 (direct acting epoxides formed after metabolic activation)“. Ein positiver Alert zeigte sich jedoch für „DNA binding by OECD“ (Michael Addition nach metabolischer Aktivierung). Damit ergab die Auswertung, dass Indan und seine Metaboliten gleich beurteilt werden. Inden zeigte dagegen für diesen (und auch alle anderen) Strukturparamater keine positiven Alerts.
3.9.5 SCHLUSSFOLGERUNG
Die Datenlage zu Indan ist ungenügend für eine stoffspezifische Bewertung.
In der Strukturanalyse mittels der OECD Toolbox ergab sich ein einziger struktureller Alert für potenzielle Gentoxizität. Bei dieser Sachlage wird bei fehlenden stoffspezifischen Daten hilfsweise eine Bewertung auf Basis des TTC-Ansatzes für gentoxische Substanzen vorgeschlagen. Dies würde eine Geringfügigkeitsschwelle von 75 ng/L bedeuten (0,15 µg/Person und Tag unter Berücksichtigung von 2 L Trinkwasseraufnahme). Alternativ könnte unter humantoxikologischen Aspekten der praktisch identische „Gesundheitliche Orientierungswert“ (GOW) von 0,1 µg/Liter für nicht stark gentoxische Substanzen (LAWA, 2004) verwendet werden. Einer Übernahme der Geringfügigkeitsschwelle des strukturverwandten Indens (möglich aufgrund ausreichend stoffbezogener Daten) steht ohne weitere experimentelle Daten entgegen, dass nur für Indan, nicht aber Inden, ein positiver Alert für potenzielle Gentoxizität existiert. Bei ausreichender Absicherung fehlender Gentoxizität des Indans wäre dagegen eine solche Übernahme in Betracht zu ziehen.
In die Bewertung nicht einbezogen werden kann der Endpunkt endokrine Wirkungen: der Indan-Metabolit Indanol zeigte im CALUX-Assay (Brinkmann et al., 2014) östrogene Aktivität. Indan selbst wurde nicht getestet, aber die in vitro-Befunde zu Indanol ergeben Verdachtsmomente auf endokrine Wirkungen auch für die Muttersubstanz, welche aber mangels geeigneter in vivo-Daten noch nicht in die Bewertung einfließen können.

The committee did not find human data on the effects of indene
following exposure by inhalation. Oxidation of indene either or not
followed by glutathione conjugation, has been demonstrated to be a
major route of detoxification in rats and guinea pigs. In guinea pigs, up to
9% of an intraperitoneally administered dose of indene was excreted
within 24 hours as the metabolite 2-hydroxy-l-methylthioindane. In rats,
urinary excretion of mercapturic acids of indene has been described.
Indene was not irritating to the skin of rats and guinea pigs. The
committee did not find data on the potential eye irritating or sensitising
properties of indene. Indene does not need classification on the basis of
its acute inhalation toxicity (4-hour LC50: >5000 mg/m3) or acute oral
toxicity (LD50 rat: 2300 mg/kg). There are no data available on acute
dermal toxicity of indene. indene was negative in mutagenicity, tests in
S. typhimurium strains TA98, TA100, TA1535, and TA1537 with and
without metabolic activation. The committee did not find adequate data
on toxicity following intermittent repeated exposure by inhalation nor on
carcinogenicity or reproduction toxicity. The committee considers the
toxicological data base on indene too poor to justify recommendation of
a health-based occupational exposure limit. The committee concludes
that there is insufficient information to comment on the level of the
present MAC value.

Anhand der vorliegenden stoffspezifischen Daten kann auf Basis der Studie mit wiederholter Exposition eine Geringfügigkeitsschwelle abgeleitet werden. Für eine fundierte Bewertung muss aber eine Übersetzung aus dem Japanischen erfolgen.

This standard evaluation procedure is part of the quality assurance programme installed at FoBiG following principles as outlined by Malmfors et al. (2004) (Tox. Letters, 35, 19-23) for Good Evaluation Practice.
The SEP describes principles for performing data searches for the purpose of working out a technical dossier under Regulation (EC) No 1907/2006 (REACH)
2 Database searches
Relevance of hits found in databases depends on the parts of the technical dossier to be worked out (see Terms of Reference in the Final Evaluation Report). In bibliographic databases relevance is scrutinized generally based on contents of titles and abstracts. If relevance of a document remains unclear after this step, the original document is ordered.
2.1 First step: publicly available databases
1 Identity of the substance is verified by a search using the database CHEMIDplus at http://toxnet.nlm.nih.gov/
2 Entries in Toxline and other bibliographic databases at TOXNET are searched by checking titles and abstracts for relevant data.
3 Pubmed (Medline) is searched at http://www.ncbi.nlm.nih.gov by checking titles and abstracts for relevant data. To this end, CAS numbers and suitable names (chemical names, trade names, colour index names or others) are used as search terms. Suitable terms have to be selected case by case and documented.
2.2 Second step: internet search
1. Again, CHEMIDplus at http://toxnet.nlm.nih.gov/ is used as a starting point.
2 Selected internet locators identified in CHEMIDplus containing information on the substance are checked for relevance.

4 A search strategy is developed based on the results of the searches in Toxline and Pubmed. A superbase search (searching simultaneously in several databases) is employed. Hits are checked for doublets and relevant bibliographic data are ordered electronically.
5 Based on contents of titles/abstracts hits are screened and documents considered relevant are retrieved in original by bibliographic services.
2.4 Fourth step: databases at STN
1 A search for relevant toxicological and ecotoxicological data is performed in Chemical Abstracts at STN-Karlsruhe (STN/ZCAplus).
2 A search strategy is developed based on the results of previous searches. Typically a search using roles (“adverse effects”) is combined with the registry number.
6 Based on contents of abstracts hits are screened and documents considered relevant are retrieved in original by bibliographic services.
