Zusammenfassung
In den Blutzellen unbehandelter und immunisierter Insekten wird histochemisch eine steigerungs
fahige NADH- bzw, NADPH-Oxidase-Aktivitat nachgewiesen, die nach KLEBANOFF uber die H 202
Bildung an dem Zustandekommen einer intrazellularen Bakterizidie beteiligt ist.
Summary
In the hemocytes of a control group and a group of immunised insects an onhanceable NADH
or NADPH-oxidase-activity is demonstrated histochemically which, according to KLEBANOFF, hasa share in the development of an intracellular antibacterial activity via the synthesis of H 202•
1. Einleitung
Nachdem KLEBANOFF (1967 a, b, 1968, 1970, 1974a, b) und KLEBANOFF u. a. (1965,1966,1972) zeigen konnten, daB die wahrend der Phagozytose in menschlichen Leukozyten auftretende Bakterizidie auf ein Zusammenwirken von Peroxidase, H 20 z undHalogen-lonen beruht, konnte dieser Wirkungsmechanismus durch andere Autorenund auch bei weiteren Wirbeltieren nachgewiesen werden (ALLEN 1975a, b, ALLENet al. 1974; BABIOR et al. 1973, 1975; BHUYAN et al. 1974; BORDER et al. 1974;DRUTZ et al. 1974; HOMAN-MULLER et al. 1975; JACOBS et al. 1973; LAWRENCE et al.1971; MANDELL 1974, MANDELL et al. 1969, McRIPLEY et al. 1967 a, b; ODEBERG et al.1975; OLSSON et al. 1972; PINCUS et al. 1971; SIMMONS et al. 1973; SIMON et al. 1971;SOLBERG et al. 1973; SOUILLET et al. 1974; STRAUSS et al. 1971; TERITO et al. 1974;
VILDE 1974; WELSH et al. 1971). Wahrend der Phagozytose kommt es in den Leukocyten der untersuchten Sauger zu einer deutlichen Stoffwechselanderung: erhohteGlykolyse, gesteigerter 02-Verbrauch und gesteigerte H 20 2- Produktion. Zugleich er-

Der Vorgang der Phagozytose ist eine bei fast all en Tieren vorkommende Erscheinung, die einerseit s eine nutritive und andererseits eine prot ektive Funktion zu erfiillen hat . Es ist daher nicht unwahrscheinlich, daB auch bei anderen Tiergruppen wahre nd der Phagozytose die gleichen oder ahnliohen Stoffwechselprozesse ablaufen, wieman sie fur die Leukozyten der Sanger nachweisen konnte.
ANDERSON et al. (1973) und ANDERSON (1974) untersuchten als erste den Stoffwechsel von Insektenblutzellen wahrend der Phagozyt ose, und sie fanden bei BIabera eineErhohung des 02-Verbrauchs, der Glukose-Oxidation und der Glykolyse. Es gela ngihnen jedo ch ni eht, di e zur Bildung von H 202 notwendige NADH- oder NADPH-Oxidase nachzuweisen.
Die vorliegende Arbeit bringt den histo chemischen Na chweis einer NADH - bz w,NA DP H -Oxidase -Aktiv itiit in Insektenblutzellen , die sich durch Phagozytose deutlich steigern liiBt.

2. Material und Methode

3. Ergebnisse
3.1. Nawphoita cinerea
In den unbehandelten B lutproben lassen sich 2 Blutzelltypen mit NADH -OxidascAktivit iit deutlich unt erscheiden:

a) einige groBe Zellen mit groBem, gut ab gegrenztem Pl asmasaum (Abb. 1, AZellen).
b) zahlreiche kleinere Zellen mit groBem Kern und schma lem nicht sehr deutlichabgrcnzbarem Plasmasaum (Abb. 1, B-ZelIen).
In den mit Tusche behandelten Blutproben ist der Anteil der B-Zellen groBer geworden . Im Plasma dieser Zellen erscheinen jetzt randstandig Vakuolen und deutlicher gefarbt e Granula (Abb. 2).
3.2. A cheta domesticus
Die unbehandelten Biutproben.zeigen positi v gefiirb te H iimozyten mit groBem K ernund einem schmalen , glat te n Plasmasaum, Nur weni ge ZelIen mit breiterem Plasmasaum besitzen eine NADH-Oxidase-Akti vitiit (Abb. 3).

In den behandelten Bluiproben ist del' Anteil del' gefiirbten Blutzellen nicht nurgrofser, sondern auch die schmalen Plasmasiiume diesel' Zellen sind deutlich vakuolisiert (Abb. 4).
3.3. Galleria mellonella
In den unbehandelten Kontrollpriiparaten sind vereinzelte, groBvakuolisierte Degenerationszellen (MOHRIG u. a. 1970) mit blaugefiirbten Granula im sparsamen peripheren Plasma oder um den Kern herum versehen.
Nur ganz wenige Plasmatocyten zeigen im Zellplasma eine positive Farbreaktion.Die Blutproben behandelter Gallerialarven zeigen gegeniiber del' Kontrolle deutliche
Veriinderungen:Die Degenerationszellen sind verschwunden, und dafur sind zahlreiche, kleine Pro
hamozyten im Plasma granular gefarbt, Diese Prohamozyten oder jungen Plasmatocyten sind groBtenteils randstandig vakuolisiert (Abb. 5).

4. Diskussion
