adenocarcinoma of thestomach or GEJ• HER2-positive tumor defined as either IHC 3+ or IHC 2+,

Multizentrische Phase II Studie
Haupteinschlusskriterien• Patienten mit metastasiertem,
histologisch gesichertem, kolorektalem Karzinom, UICC Stadium IV
• Oligometastasierte Erkrankung, definiert als mindestens 1 messbare Läsion größer als 1cm (RECIST v1.1) und maximal 5 Läsionen an maximal 3 Lokalisationen
• Patienten, die nach 3-6 Monaten Induktionschemotherapie (Irinotecan- oder Oxaliplatin-haltige Kombinationschemotherapie mit Bevacizumab) weder eine progrediente noch eine resektable Tumorerkrankung zeigen.
• Maximale Therapieunterbrechung vor Beginn der Studientherapie und nach Beendigung der Induktionstherapie von 6 Wochen.
• ECOG ≤ 1, Lebenserwartung > 3 Monate

Einschlusskriterien• Metastatic or unresectable locally advanced/recurrent breast cancer HER2-positive
disease documented as ISH-positive and/or 3+ by IHC on previously collected tumor tissue and prospectively confirmed central laboratory prior to study enrollment. Tumorgewebe muss vorhanden sein.
• Disease progression on the last regimen received as defined by the investigator.• Prior treatment with an anthracycline, trastuzumab, a taxane, lapatinib, and capecitabine
in the neoadjuvant, adjuvant, locally advanced, or recurrent/metastatic setting and documented disease progression (by investigator assessment) after at least two regimens of HER2-directed therapy in the metastatic or unresectable locally advanced/recurrent setting.
• Patients must have had at least 6 weeks of prior exposure in the metastatic (or unresectable locally advanced/recurrent) setting to lapatinib and capecitabine (given together or separately) unless they were intolerant of lapatinib and/or capecitabine. Patients who were found to be intolerant to lapatinib can be considered eligible if they experienced disease progression during a single trastuzumab-based regimen in the metastatic (or unresectable locally advanced/recurrent) setting.

Bestimmung des EGFR Mutationsstatus anhand einer Tumorprobe

Einschlusskriterien
• Histologisch oder zytologisch bestätigtes lokal fortgeschrittenes (Stadium IIIA/B) nicht kurativ behandelbares oder metastasiertes NSCLC
• Neu diagnostizierte Patienten mit lokal fortgeschrittenem oder metastasiertem NSCLC die bisher keine systemische Therapie erhalten haben oder Patienten, die vorher eine adjuvante Chemotherapie erhalten haben (außer: EGFR-TKI)
• Vorhandensein eines Tumorblocks oder einer zytologischen Probe zum Zeitpunkt des Einschlusses
Ausschlusskriterien
• Scherwiegende oder unkontrollierbare systemische Erkrankung (z.B. instabile oder nicht kompensierte respiratorische, kardiologische, hepatische oder renale Erkrankung)
• Vorhandensein einer anderen signifikanten klinischen Erkrankung oder Laborbefunde die eine Studienteilnahme für diesen Patienten nicht wünschenswert erscheinen lassen.
